本研究用72只新西兰家兔,观察了柴葛解肌汤(No.Ⅰ)和新加香薷饮(No.Ⅱ)两种清热方剂对家兔白细胞致热原(leucocytic pyrogen,LP)性发热效应及脑脊液cAMP含量影响的比较。结果表明:(1)两种方剂灌胃对家兔正常体温没有明显影响(P>0.05)。(2)单独注射LP引起直肠温度上升0.90±0.12℃,静注LP前一小时灌服No.Ⅰ或No.Ⅱ方剂直肠温度上升分别为0.83±0.19和0.45±0.15(△T℃),与单独注射LP组比较有非常显著性差异(P<0.01),说明这两种方剂增有明显的解热作用。(3)静注LP后一小时检测家兔脑脊液cAMP含量为140.94±51.74pmol/ml,而No.Ⅰ+LP组脑脊液cAMP含量则为83.48±8.11pmol/ml。两者差异显著(P<0.05),表明No.Ⅰ方剂不仅抑制LP引起的发热效应,还明显降低LP引起的脑脊液的cAMP含量增高。但No.Ⅱ+LP组同时点的cAMP含量为114.48±11.93pmol/ml,与单独注射LP组比较差异不显著(P>0.05),表明No.Ⅱ方剂虽能抑制LP引起的发热效应,但不能明显降低LP引起的脑脊液的cAMP含量升高。根据这些结果,作者推论:《柴葛解肌汤》可能主要通过降低脑内cAMP含量的上升取得解热效应,而《新加香薷饮》则可能通过另外的方式而导致解热。
In this study, 72 New Zealand rabbits were used to observe the effects of heat-clearing formulas of Chaige Jieji Tang (No.I) and Xinjiaxiangxiyin (No.II) on leucocytic pyrogen (LP) in rabbits. Comparison of thermal effects and cAMP content in cerebrospinal fluid. The results showed that: (1) There was no significant difference in normal body temperature between rabbits fed with the two formulas (P>0.05). (2) The rectal temperature increased by 0.90±0.12°C caused by LP injection alone, and the rectal temperature increased by 0.83±0.19 and 0.45±0.15(△T°C), respectively, in the No.I or No. II prescription fed one hour before intravenous injection of LP. There was a significant difference (P<0.01) between injections of LP alone, indicating that these two formulas had significant antipyretic effects. (3) The cAMP content of rabbit cerebrospinal fluid was 140.94±51.74pmol/ml one hour after intravenous injection of LP, while cAMP content in cerebrospinal fluid of No.I+LP group was 83.48±8.11pmol/ml. There was a significant difference between the two groups (P<0.05), indicating that the No. I prescription not only inhibited the fever effect caused by LP, but also significantly reduced the cAMP content of cerebrospinal fluid caused by LP. However, at the same time, the cAMP content in the No.II+LP group was 114.48±11.93 pmol/ml, which was not significantly different from that of the LP group alone (P>0.05), indicating that the No.II prescription can suppress the heat effect caused by LP. Can not significantly reduce the LP-induced increase in cAMP content of cerebrospinal fluid. Based on these results, the authors deduced that “Chaige Jieji Decoction” may achieve the antipyretic effect mainly by lowering the increase of cAMP content in the brain, while “Xinjiaxiang Yinchu” may lead to antipyretic effects in another way.
