目的探讨天然脑活素对复制性衰老人胚肺二倍体成纤维细胞(MRC-5)端粒酶活性的影响,以阐明其延缓衰老的作用及分子机制。方法镜下观察细胞形态变化,绘制细胞生长曲线,MTT法检测细胞增殖活度,TRAP-ELISA方法检测细胞端粒酶活性的变化。结果天然脑活素孵育的MRC-5细胞老龄化不明显,细胞增殖能力较空白老龄对照组明显增加(P<0.05);天然脑活素处理的细胞端粒酶活性较空白老龄对照组明显上调(P<0.05)。结论天然脑活素可能通过促进细胞增殖,激活细胞端粒酶的表达,进而有效地延缓细胞的复制性衰老。
Objective To investigate the effect of natural cerebrolysin on the telomerase activity of human diploid human embryo lung diploid fibroblasts (MRC-5) in order to elucidate its anti-aging effect and its molecular mechanism. Methods Morphological changes were observed microscopically, cell growth curve was drawn, cell proliferation activity was detected by MTT assay, and telomerase activity was detected by TRAP-ELISA. Results The MRC-5 cells incubated with natural cerebrolysin had no obvious aging, and the cell proliferation ability was significantly increased compared with the blank control group (P <0.05). The activity of telomerase in natural cerebrolysin group was significantly higher than that of the blank control group (P <0.05). Conclusions Natural brain activin may effectively delay the replication of cells by promoting the cell proliferation and activating the expression of telomerase.
