本课题研究骨形成蛋白(BMP)和微血管生成与骨肉瘤发展的关系.应用免疫组化SP法,对40例骨肉瘤标本进行抗BMP多克隆抗体和八因子相关抗原单克隆抗体(FvmRAg)免疫组化染色,检测骨肉瘤组织和微血管密度(MVD).本实验结果显示,BMP的表达主要见于肿瘤细胞浆内,呈较均匀的棕色颗粒状,骨基质着色也较深.BMP的表达具有明显的异质性,在肿瘤性骨样组织幼稚区,肿瘤浸润前缘和新生毛细血管附近BMP表达很强.40例骨肉瘤标本中,36例阳性,阳性率为90%.BMP的表达和肿瘤体积、Dahlin’s分型和Price’s分级无关(P>0.05),但和血道转移、软组织浸润有关(P<0.05).对骨肉瘤血管的研究显示,MVD亦呈质性,肿瘤周围的微血管较肿瘤内部密集,尤其是肿瘤浸润前缘可见簇状的微血管,血管形态不规整,分布不均匀.同样,伴有软组织浸润和血道转移的MVD明显高于不伴软组织浸润和血道转移组(P<0.05).本研究显示BMP对骨肉瘤的血管生成具有一定的影响,在骨肉瘤BMP高表达组,MVD显著高于低表达组(P<0.05),表现出BMP和MVD的
This study investigated the relationship between bone morphogenetic protein (BMP) and angiogenesis and the development of osteosarcoma.Immunohistochemical SP method was used to immunize anti-BMP polyclonal antibody and FvmRAg antibody against 40 osteosarcoma specimens The results of the present study showed that the expression of BMP was mainly found in the cytoplasm of tumor cells, with a relatively uniform brown granular shape and a deep staining of bone matrix.The expression of BMP was obviously , And there was a strong expression of BMP in the tumor-like tissue infiltration zone, tumor infiltration margin and nascent capillaries.Among 40 osteosarcoma specimens, 36 were positive and the positive rate was 90% .BMP expression and tumor Volume, Dahlin’s classification and Price’s classification (P> 0.05), but not with Price’s classification (P> 0.05), but was related to hematogenous metastasis and soft tissue infiltration (P <0.05) .Among the study on osteosarcoma, MVD was also qualitative. Clustered microvessels were found densely, especially in the anterior margin of tumor infiltration with irregular shape and uneven distribution of blood vessels. Similarly, MVD with soft tissue infiltration and hematogenous metastasis was significantly higher than that without soft tissue infiltration and hematogenous metastasis (P <0.05) .this Studies show BMP generated has a certain influence on vascular osteosarcoma, osteosarcoma BMP in high expression, MVD was significantly higher than the low expression group (P <0.05), and MVD showed BMP
