目的研究运动前补充罗汉果槲皮素对力竭运动大鼠肝组织抗氧化能力和肝细胞凋亡调控基因Bcl-2和Bax表达的影响。方法选用SD雄性大鼠30只,随机分为3组,每组10只,即安静对照组(C)、力竭训练组(E)和力竭训练+罗汉果槲皮素补充组(HE)。大鼠于每天上午8:00进行负重游泳力竭训练,1次/d,6 d/周,持续训练5周。5周力竭训练结束后,宰杀大鼠。根据试剂盒的方法测定大鼠肝组织SOD、CAT、GSH-Px活性和MDA含量并且通过逆转录聚合酶链式反应(RT-PCR)测定肝细胞凋亡调控基因Bcl-2和Bax的表达。结果大鼠力竭运动前灌胃槲皮素对大鼠肝组织SOD、CAT、GSH-Px活性有明显的升高作用,MDA含量有所下降。力竭游泳训练促进大鼠肝组织细胞调亡基因Bcl-2表达下调而Bax表达升高,力竭游泳运动前补充罗汉果槲皮素促进肝组织细胞凋亡调控基因Bcl-2高表达而Bax表达则下调。结论补充罗汉果槲皮素对力竭运动大鼠有抗自由基氧化的功能,可减轻脂质过氧化反应和抑制肝细胞的调亡,从而起到保护肝脏的作用。
Objective To study the effect of supplementation of mogroside quercetin on the anti-oxidation ability and expression of hepatocyte apoptosis regulatory gene Bcl-2 and Bax in rats after exhaustive exercise. Methods Thirty male Sprague-Dawley rats were randomly divided into 3 groups with 10 rats in each group, namely, the quiet control group (C), the exhaustive training group (E) and the exhausted training group and the mogroside quercetin supplemented group (HE). Rats performed exhausted swimming training every day at 8:00 am, once/d, 6 d/week, and continued training for 5 weeks. After 5 weeks of exhausted training, the rats were slaughtered. The activity of SOD, CAT, GSH-Px and MDA in liver tissue of rats were determined according to the kit method. The expressions of Bcl-2 and Bax, the apoptosis-regulatory genes of hepatocytes, were determined by reverse transcription polymerase chain reaction (RT-PCR). Results The contents of SOD, CAT and GSH-Px in the liver tissue of rats were significantly increased by quercetin pretreatment before exhaustive exercise. The content of MDA decreased. Exhaustive swimming training promoted down-regulation of Bcl-2 expression and increased expression of Bax in liver tissue of rat livers. Exogenous swimming exercise supplemented mogroside quercetin to promote liver cell apoptosis and high expression of Bcl-2 and Bax expression. Then down. Conclusions Supplementation of mogroside quercetin has anti-free radical oxidation effect on exhausted rats, which can reduce the lipid peroxidation and inhibit the apoptosis of hepatocytes, thus protecting the liver.
