目的探讨大鼠实验性胃溃疡自愈期间下颌下腺三叶因子1(TFF1)的表达变化。方法采用免疫组织化学和RT-PCR法,分别从蛋白水平及基因水平检测42只溃疡组、21只盐水组和6只正常组大鼠下颌下腺TFF1的表达。结果免疫组织化学SP法显示,正常大鼠下颌下腺颗粒曲管的多颗粒细胞呈TFF1免疫反应阳性,各级导管管腔内也有TFF1阳性物质,纹状管管腔游离面可见线条状TFF1阳性物。溃疡组下颌下腺TFF1肽积分吸光度值明显增加,高于相应的盐水对照组和正常组(P<0.05或P<0.01),其中1d、2d、4d、6d TFF1肽积分吸光度值逐渐增加,6d最高,10d、14d、23d也显著高于盐水对照组。RT-PCR结果显示,下颌下腺有TFF1 mRNA转录,且溃疡组TFF1/GAPDH mRNA吸光度比值在溃疡2~23d均高于盐水对照组和正常组(P<0.01或P<0.05)。结论下颌下腺TFF1肽在大鼠实验性胃溃疡形成和愈合过程中高表达,主要通过导管系统排泄,参与实验性胃溃疡的愈合。
Objective To investigate the expression of trefoil factor 1 (TFF1) in experimental rat gastric ulcer during healing. Methods Immunohistochemistry and RT-PCR were used to detect the expression of TFF1 in submandibular gland of 42 ulcer rats, 21 saline rats and 6 normal rats at protein and gene level respectively. Results Immunohistochemical SP method showed that TGF-1 immunoreactivity was positive in multi-granule cells in normal rat submandibular gland curvatures, and TFF1-positive material was also present in the lumen of the catheters at all levels. . The absorbance value of TFF1 peptide in ulcer group was significantly higher than that in saline control group and normal group (P <0.05 or P <0.01), and the absorbance value of TFF1 peptide in 1d, 2d, 4d and 6d gradually increased, , 10d, 14d, 23d also significantly higher than saline control group. RT-PCR results showed that TFF1 mRNA was transcribed in submandibular gland, and the ratio of TFF1 / GAPDH mRNA absorbance in ulcer group was higher than saline control group and normal group (P <0.01 or P <0.05) at 2 ~ 23 d after ulcer. Conclusion TFF1 peptide in the submandibular gland is highly expressed in the process of experimental gastric ulcer formation and healing in rats. It is mainly excreted through the catheter system and participates in the healing of experimental gastric ulcer.
