用旋毛虫感染猪血清对旋毛虫肌幼虫cDNA文库进行了免疫筛选,从1 6×105个重组噬菌体中筛选出21个阳性克隆。阳性克隆的测序结果表明:有9个克隆为未曾报道过的新基因;有9个克隆不含有开放阅读框架(ORF),与旋毛虫线粒体DNA同源,编码核糖体大亚基RNA;3个克隆为旋毛虫已知基因,其中克隆ML12,34与Serineproteaseinhibitor[Trichinellaspiralis](AAF63473)同源性为99%,克隆ML42与HypotheticalOFR17 20[Trichinellaspiralis](AAB48489)同源性为99%,与21kDExcretory secretoryprotein[Trichinellapseudospiralis](AAF79206)同源性为90%,这为进一步研究基因重组抗原奠定了基础。
Trichinella spiralis larvae cDNA library was immunologically screened with Trichinella inoculated swine serum and 21 positive clones were screened from 16 × 105 recombinant phage. The sequencing results of positive clones showed that 9 clones were not reported as new genes, 9 clones did not contain open reading frame (ORF), homologous to Trichinella mitochondrial DNA and encoded ribosomal large subunit RNA, 3 Clone ML12,34 has 99% homology with Serine protease inhibitor [Trichinellalaspiralis] (AAF63473), and clone ML42 has 99% identity with Hypothetical OF 1717 [Ach4841] (AAB48489), and 21 kD Excretory secretory protein [ Trichinella pseudospiralis] (AAF79206) with a homology of 90%, which laid the foundation for further study of recombinant antigen.
