目的:研究白细胞介素-(IL)1β多克隆抗体对新生大鼠缺氧缺血性脑损伤(HIBD)后对损伤大脑的保护效应。方法:制备左脑HIBD的新生大鼠模型,于HIBD前后分别腹腔注射一定浓度的IL-1β多克隆抗体,观察用药前后损伤大脑脑水含量及组织病理学变化。结果:HIBD前后腹腔注射IL-1β多克隆抗体可明显降低脑水含量(P<0.01),但对其相应mRNA转录无影响。组织病理学观察发现用药组大脑水肿明显减轻,神经元死亡明显减少。结论:IL-1β多克隆抗体可以拮抗IL-1β的炎性作用,对HIBD大脑有保护效应。
Objective: To investigate the protective effects of polyclonal antibody against interleukin - 1 (IL - 1β) on injured brain in neonatal rats after hypoxic - ischemic brain damage (HIBD). Methods: A neonatal rat model of left HIBD was prepared. The polyclonal antibody against IL-1β was injected intraperitoneally before and after HIBD. The changes of brain water content and histopathological changes were observed before and after treatment. Results: The intraperitoneal injection of IL-1β polyclonal antibody before and after HIBD significantly decreased the content of brain water (P <0.01), but had no effect on the corresponding mRNA transcription. Histopathological observation showed that the edema of the brain in the treatment group was significantly reduced and the neuronal death was significantly reduced. Conclusion: The polyclonal antibody of IL-1β can antagonize the inflammatory effect of IL-1β and protect the brain of HIBD.
