目的:建立HPLC-PDA多波长检测法同时测定乌梅肉中柠檬酸,5-羟甲基糠醛(5-HMF),新绿原酸的含量。方法:采用XBridgeTMC18色谱柱(4.6 mm×250 mm,5μm),以(A)0.5%磷酸二氢铵溶液(p H 3.0)-(B)乙腈为流动相进行梯度洗脱(0~10 min,95%~80%A;10~15 min,80%~70%A;15~16 min,70%~95%A;16~21 min,95%A),流速0.8 m L·min-1,柱温25℃,检测波长分别为210,284,327 nm。结果:通过对线性关系的考察,乌梅肉中柠檬酸,5-HMF,新绿原酸分别在63.30~8 100.00 mg·L~(-1)(r=0.999 9),0.50~32.00 mg·L~(-1)(r=0.999 9),0.19~12.00 mg·L~(-1)(r=0.999 8)内线性良好,各成分的回收率在97.21%~101.01%,RSD均<2.0%,供试品溶液中3种指标性成分至少在24 h内稳定性良好。在不同产地乌梅肉样品中柠檬酸的平均质量分数为318.71 mg·g-1,5-HMF的平均质量分数为1.99 mg·g-1,新绿原酸的平均质量分数为2.01 mg·g-1,以浙江和四川产的乌梅质量最好。结论:该方法准确、快速、成本较低,可用于乌梅肉中这3个成分的含量测定,为乌梅在食品和药品开发的质量控制方面提供了参考。
Objective: To establish a HPLC-PDA multi-wavelength detection method for simultaneous determination of citric acid, 5-hydroxymethylfurfural (5-HMF) and chlorogenic acid in ebony meat. METHODS: A gradient elution was performed on a XBridge TMMC18 column (4.6 mm × 250 mm, 5 μm) with (A) 0.5% ammonium dihydrogen phosphate solution (p H 3.0) - (B) 95% A, 10-15 min, 80-70% A, 15-16 min, 70-95% A, flow rate 0.8 m L · min-1, The column temperature was 25 ℃, the detection wavelength was 210,284,327 nm. Results: The results showed that citric acid, 5-HMF and chlorogenic acid in Fructus Mume were 63.30-8.00.00 mg · L -1 (r = 0.999 9), 0.50-32.00 mg · L -1 (R = 0.999 9). The linearity was good in the range of 0.19-12.00 mg · L -1 (r = 0.999 8). The recoveries of all components ranged from 97.21% to 101.01% with RSDs less than 2.0% The test solution for the three indicators of at least 24 hours of good stability. The average mass fraction of citric acid was 318.71 mg · g-1 and the average mass fraction of fresh chlorogenic acid was 2.01 mg · g-1 , The quality of ebony produced in Zhejiang and Sichuan is the best. Conclusion: The method is accurate, rapid and low cost and can be used for the determination of the three components in the plum meat, which provides a reference for the quality control of plum in food and drug development.
