目的:建立测定滇丹参中丹酚酸B含量的HPLC法,并与丹参中丹酚酸B的含量进行比较。方法:采用Inertsil DiolC18(250mm×4.6mm,5μm)色谱柱,以甲醇-乙腈-甲酸-水(40∶20∶1∶39)为流动相,流速1.0mL.min-1,检测波长280nm。结果:水回流提取分离效果好,干扰成分少;丹酚酸在0.13~0.65μg范围内线性关系良好(r=0.999 9),平均加样回收率为98.5%;滇丹参与丹参中丹酚酸B的含量存在着较大的区别。结论:本实验建立的方法重现性好,定量准确;滇丹参与丹参水溶性主要区别成分应作进一步定性研究,其药理作用的异同亦应作深入的研究。
OBJECTIVE: To establish a HPLC method for the determination of Salvianolic acid B in Salvia miltiorrhiza Bge.and to compare the content of salvianolic acid B in Salvia miltiorrhiza. Methods: Inertsil Diol C18 (250 mm × 4.6 mm, 5 μm) column was used with mobile phase of methanol-acetonitrile-formic acid-water (40:20:1:39) as the mobile phase at a flow rate of 1.0 mL · min- Results: The water reflux extraction and separation effect was good with less interfering components. The linearity of salvianolic acid in the range of 0.13 ~ 0.65 μg was good (r = 0.999 9), the average recovery was 98.5% B content there is a big difference. Conclusion: The method established in this experiment has good reproducibility and quantitative accuracy. The main components of the water-soluble constituents of Yandan participating in Salvia miltiorrhiza should be further qualitatively studied, and the similarities and differences of the pharmacological effects should be further studied.
