目的探讨体外培养的血管平滑肌细胞(VSMC)增殖细胞核抗原(PCNA)表达对其凋亡的激光诱导率的影响。方法组织贴块法培养VSMC,饥饿法同步化,在不同的刺激因子作用之后,免疫组化法检测细胞PCNA表达率,各组细胞经510.6nm的铜蒸汽激光照射,TUNEL法记数细胞凋亡率。结果PCNA表达高的细胞,在激光的诱导下易发生凋亡;相反,PCNA表达低细胞,凋亡诱导率较低。结论经不同生长因子刺激后VSMC的PCNA表达率不同,PCNA表达率的不同对激光的凋亡诱导率产生影响。
Objective To investigate the effect of proliferating cell nuclear antigen (PCNA) expression on the rate of laser induced apoptosis in cultured vascular smooth muscle cells (VSMCs). Methods VSMCs were cultured by tissue patch method and synchronized by starvation method. The expression of PCNA was detected by immunohistochemical method after different stimulation factors. The cells were irradiated by copper vapor laser at 510.6nm and the apoptosis was counted by TUNEL method rate. Results The cells with high expression of PCNA were prone to apoptosis under laser induction. On the contrary, PCNA expressed low level of cells with low induction of apoptosis. Conclusion The expression of PCNA in VSMC after different growth factors stimulation is different. The difference of PCNA expression rate affects the induction rate of apoptosis of laser.
