目的 探讨在中国汉族哮喘家系中 ,HLA DQ基因与哮喘的相关性。方法 对 98例哮喘家系成员 ,用PCR 序列特异性引物 (PCR SSP)技术对HLA DQA1和B1进行基因分型 ,并与正常对照进行比较。结果 发现DQA1 0 10 1和DQA1 0 6 0 1等位基因频率在哮喘患者组 (4 0 0 % ,45 0 % )较正常对照组 (16 4% ,13 4% )显著升高 (χ2 =6 186 0 ,P <0 0 5 ,RR =3 39;χ2 =11 6 0 90 ,P <0 0 1,RR =5 2 7) ;DQB1 0 30 3和DQB1 0 6 0 1等位基因频率在哮喘患者组 (5 5 0 % ,47 5 % )较正常对照组 (13 7% ,13 7% )显著升高 (χ2 =15 740 0 ,P <0 0 1,RR =7 6 8;χ2 =10 930 0 ,P <0 0 1,RR =5 6 9)。同时发现HLA DQB1 0 2 0 1等位基因频率在对屋尘螨抗原特异性IgE反应哮喘家系成员 (39 4% )较家系中非特应症者 (12 0 % )显著高频表达 (t =2 382 5 ,P <0 0 5 )。结论 HLA DQA1 0 10 1, 0 6 0 1和DQB1 0 30 3, 0 6 0 1是哮喘遗传易感等位基因 ;HLA DQB1 0 2 0 1限定对屋尘螨抗原特异性IgE反应
Objective To investigate the relationship between HLA DQ gene and asthma in asthma pedigrees of Han Chinese. Methods 98 cases of asthmatic family members were genotyped by PCR SSP and HLA-DQA1 and B1, and compared with the normal control. The results showed that the allele frequencies of DQA1 0 10 1 and DQA1 0 601 were significantly higher in the asthma group (40%, 45%) than in the normal control group (16.4%, 134%) (χ2 = 6 186 0, P <0 0 5, RR 3 39, χ 2 = 11 6 0 90, P 0 01, RR 5 327). The allele frequencies of DQB1 0 30 3 and DQB1 0 601 were significantly higher in asthma The patient group (55.0%, 47.5%) was significantly higher than the normal control group (13.7%, 13.7%) (χ2 = 15 740 0, P <0.01, RR = 930 0, P <0 0 1, RR = 5 6 9). At the same time, we found that the frequency of allele HLA DQB10201 was significantly high frequency in the asthmatic pedigrees (39 4%) and non-atopic subjects (12 0%) of house dust mite antigen-specific IgE reaction (t = 2 382 5, P <0 0 5). Conclusions HLA DQA1 0 10 1, 0 6 0 1 and DQB1 0 30 3, 0 6 0 1 are susceptible alleles of asthma. HLA DQB1 0 0 1 limits the antigen-specific IgE response to house dust mites
