选用适合的冻存条件贮存肿瘤标本可为“SRC”实验中有计划顺应时机提供标本。本文介绍了采用小鼠MA737乳腺癌标本经过4℃/24小时,-20℃/72小时,-80℃/72小时冻存处理后再移植到近交系小鼠体内观察生长情况和用0.02%苔盼蓝染色计算细胞存活率的试验分析了其影响因素和与“SRC”实验结果的关系。实验结果发现新鲜肿瘤标本和经4℃/24小时冻存处理的肿瘤标本其生长情况和细胞存活率的比值均无明显差别,病理形态无变化。经-20℃/72小时和-80℃/72小时冻存处理的标本结果次之,尤其是-20℃/72小时试验组的标本病理形态观察还发现细胞肿胀、胞浆变性变化。冻存标本时加入保护剂后的细胞对0.02%苔盼蓝染色后的N/A值明显高于不加保护剂的标本。
Selecting appropriate cryopreservation conditions for storage of tumor specimens may provide a plan to comply with timing in the “SRC” experiment. This article describes the use of mouse MA737 breast cancer specimens after 4 °C / 24 hours, -20 °C / 72 hours, -80 °C / 72 hours after cryopreservation and transplantation into inbred mice to observe the growth and use of 0.02% The trypan blue staining was used to calculate the survival rate of the cells and analyzed the influence factors and the relationship with the “SRC” experimental results. The experimental results showed that there was no significant difference in the growth rate and cell survival rate between the fresh tumor specimens and the tumor specimens treated at 4°C/24 hours after cryopreservation, and the pathological morphology was unchanged. The results of specimens cryopreserved at −20° C./72 hours and −80° C./72 hours were secondary, especially the specimens in the experimental group at −20° C./72 hours. Cellular swelling and changes in cytoplasmic degeneration were also observed. The N/A values of cells stained with 0.02% Tweed blue after the protective agent was added when the specimens were frozen were significantly higher than those without the protective agent.
