目的:初步探讨氟对人成骨细胞saos-2和小鼠骨组织中caspase14表达的影响。方法:体外建立人成骨细胞染氟模型,以不加入氟入培养基者为空白对照,实验组培养基内分别含氟化钠5、10、20、40 mg/L。以氟化钠加入饮水中,建立小鼠染氟模型。应用Western blot法检测不同剂量氟(0、5、10、20、40 mg/L)对成骨细胞saos-2中caspase14的表达,再用免疫组织化学检测不同染氟剂量(0、50、100、150 mg/L)染氟小鼠骨骼上caspase14的表达情况。结果:Caspase14在成骨细胞saos-2中各实验组表达明显上调(均P<0.01),5 mg/L组表达最高。染氟小鼠模型骨组织中亦有表达,染氟组表达均高于对照组(均P<0.05),且有逐渐增高趋势。结论:Caspase14在染氟成骨细胞和骨组织中的高表达参与了氟诱导细胞凋亡的过程。
Objective: To investigate the effect of fluoride on the expression of caspase 14 in human osteoblast saos-2 and mouse bone. Methods: The model of human osteoblast staining was established in vitro. The control group was blank control without adding fluoride to the culture medium. The experimental group contained 5, 10, 20 and 40 mg / L sodium fluoride respectively. Sodium fluoride was added to drinking water to establish a mouse model of fluoride. Western blot was used to detect the expression of caspase14 in osteoblasts saos-2 with different doses of fluorine (0, 5, 10, 20 and 40 mg / L), and the levels of caspase 14 were detected by immunohistochemistry , 150 mg / L) staining of skeletal fluorosis in mice caspase14 expression. Results: Caspase14 expression was significantly up-regulated in all experimental groups (all P <0.01), and highest in 5 mg / L osteosarcoma. Fluoride-induced mouse model bone tissue expression, fluoride group were higher than the control group (all P <0.05), and a gradual increase in the trend. Conclusion: The overexpression of Caspase14 in fluoroblasts and bone tissues is involved in the process of fluorine-induced apoptosis.
