目的:对糖安胶囊中的蛋白质成分进行含量测定。方法:采用考马斯亮蓝法测定糖安胶囊中可溶性蛋白含量。结果:牛血清标准蛋白溶液标准曲线为:Y=7.862 7X+0.077 59(R2=0.992 97),其在0.01~0.1mg/mL浓度范围内与吸光度呈良好的线性关系;样品溶液精密度RSD为1.64%;稳定性RSD为2.77%,样品溶液在60min内稳定;样品溶液中可溶性蛋白含量均值为13.4mg/g,RSD为2.36%;加样回收测得平均回收率为99.5%,RSD为2.60%;三批样品(150701、150702、150703)的可溶性蛋白含量均值为13.1mg/g,RSD为3.17%,拟定糖安胶囊中可溶性蛋白含量不得少于6.0mg/g。结论:该方法用于糖安胶囊中的蛋白质含量测定准确、可靠,可对糖安胶囊进行质量控制。
Objective: To determine the content of protein in Tangan capsule. Methods: The content of soluble protein in Tangan capsule was determined by Coomassie brilliant blue method. Results: The calibration curve of standard protein solution of bovine serum was as follows: Y = 7.862 7X + 0.077 59 (R2 = 0.992 97), which showed a good linear relationship with absorbance in the concentration range of 0.01-0.1 mg / mL. The RSD 1.64%; the RSD of stability was 2.77%; the sample solution was stable within 60 min; the average content of soluble protein in the sample solution was 13.4 mg / g and the RSD was 2.36%; the average recovery was 99.5% with RSD of 2.60 %. The average content of soluble protein in three batches of samples (150701,150702,150703) was 13.1mg / g and the RSD was 3.17%. The content of soluble protein in the preparation of Tangan Capsule should not be less than 6.0mg / g. Conclusion: This method is accurate and reliable for the determination of protein content in Tangan Capsules, which can control the quality of Tanguan Capsules.
