研究半边旗(Pteris semipinnata L, PsL)有效成分5F,6F,A对HL-60细胞DNA拓扑异构酶活性和细胞周期的影响.方法:应用pBR322质粒DNA作为底物测定酶的活性;细胞周期用流式细胞仪测定;应用噻唑蓝(MIT)法测定药物对细胞生长的抑制率.结果:5F,6F,A均能够抑制DNA拓扑异构酶Ⅰ,Ⅱ的活性,化合物6F作用细胞24 h后,可升高 S期和 G2/M期细胞,同时降低G0/G1期细胞,低浓度6F(57.8和 115.6nmol·L-1)与金雀异黄素(Gen)合用可增强它对HL-60细胞的杀伤作用, q>1.15.结论: 5F, 6F和 A明显抑制HL-60细胞DNA拓扑异构酶的活性;6F阻断细胞于 G2/M期,增强Gen对 HL-60细胞的杀伤作用。
To study the effect of 5F, 6F, A active ingredient Pteris semipinnata L (PsL) on DNA topoisomerase activity and cell cycle in HL-60 cells. Methods: Enzyme activity was determined by using pBR322 plasmid DNA as a substrate. The cell cycle was measured by flow cytometry. The inhibitory rate of the drug on cell growth was determined by MTT assay. Results: 5F, 6F and A inhibited the activity of DNA topoisomerase Ⅰ and Ⅱ. After treated with compound 6F for 24 h, the cells in S phase and G2 / M phase increased and the cells in G0 / G1 phase decreased The combined effect of 6F (57.8 and 115.6nmol·L-1) and genistein (Gen) could enhance the killing effect on HL-60 cells with q> 1.15. CONCLUSION: 5F, 6F and A significantly inhibit the DNA topoisomerase activity of HL-60 cells. 6F blocks G2 / M phase and enhances the killing effect of Gen on HL-60 cells.
