目的:分析C57 TgN(HBVadr2.0)SMMU“3号”品系转基因小鼠血清和组织中病毒蛋白的表达及其特征。方法:以138只F6~F17代C57 TgN(HBVadr2.0)SMMU“3号”品系的转基因小鼠为研究对象,采用血清ELISA检测和免疫组织化学的方法分析病毒蛋白在转基因小鼠中的表达。结果:转基因小鼠血清中HBsAg和HBeAg的阳性率分别为55.18%和25.00%,血清中HBsAg的出现时间多在3个月龄之前(占93.33%),且持续时间在9 个月以上(占95.56%),个体之间具有一定差异。转基因小鼠肝组织中至少可检测到一种病毒蛋白,HBsAg 、HBcAg和X蛋白的检出率分别为85.82%、58.24%和49.61%。HBsAg分布于肝细胞质中,HBcAg和X蛋白既可分布于肝细胞核中,也可分布于肝细胞质中。HBsAg的表达具有细胞特异性(门管区周围的肝细胞)和组织特异性(肝和肾),X蛋白除在肝脏和肾脏中表达外,还可在脑组织中表达。结论:C57 TgN(HBVadr2.0)SMMU“3号”品系转基因小鼠血清和肝肾等组织中有病毒蛋白的表达,病毒蛋白的分布具有组织细胞特异性和个体差异。
OBJECTIVE: To analyze the expression and characteristics of viral proteins in the serum and tissues of C57 TgN (HBVadr2.0) SMMU “No. 3” transgenic mice. Methods: 138 transgenic mice with the F6 ~ F17 generation of C57 TgN (HBVadr2.0) SMMU “3” strain were selected as experimental subjects. The expression of viral proteins in transgenic mice was analyzed by ELISA and immunohistochemistry . Results: The positive rates of HBsAg and HBeAg in serum of transgenic mice were 55.18% and 25.00%, respectively, and the occurrence time of HBsAg in serum was more than 3 months old (93.33%) and the duration of HBsAg was more than 9 months 95.56%), there is a certain difference between individuals. At least one viral protein was detected in the liver tissue of transgenic mice. The detection rates of HBsAg, HBcAg and X protein were 85.82%, 58.24% and 49.61% respectively. HBsAg is distributed in the liver cytoplasm, HBcAg and X protein can be distributed in the liver cell nucleus, can also be distributed in the liver cytoplasm. The expression of HBsAg is cell-specific (hepatocytes around the portal area) and tissue-specific (liver and kidney). In addition to its expression in liver and kidney, X protein is also expressed in brain tissue. Conclusion: The expression of viral proteins in the serum, liver, kidney and other tissues of C57 TgN (HBVadr2.0) SMMU “3” transgenic mice is characterized by histiocytic specificity and individual differences.
