[目的]研究亚慢性砷暴露对小鼠大脑组织神经递质代谢酶基因及其受体基因表达谱的影响。[方法]小鼠按体重随机分为3组,每组2只:4mg/L三氧化二砷(As2O3)染毒组、1mg/L As2O3染毒组和生理盐水对照组。染毒组小鼠饮用含As2O3自来水,对照组饮用0.9%生理盐水,各组小鼠每天消耗水量约20mL,均连续饮用60d,之后对其大脑组织进行基因芯片研究。[结果]与对照组比较,染砷组小鼠大脑组织的基因Th、Dbh、Tph1、Drd3、Drd4、Adra1a、Adra2a、Adra2b和Adrb3表达下调,而基因Htr1f、Htr4、Htr7表达上调。[结论]As2O3可能导致小鼠大脑组织部分神经递质合成和分解代谢酶基因表达下调,且干扰其部分受体基因的表达。
[Objective] To investigate the effects of subchronic arsenic exposure on the gene expression profile of neurotransmitter metabolic enzymes and its receptor in mouse brain. [Methods] The mice were randomly divided into 3 groups according to body weight, with 2 rats in each group: 4mg / L As2O3, 1mg / L As2O3 exposure group and saline control group. The mice in the exposure group received drinking water containing As2O3 and 0.9% saline in the control group. Each mouse consumed about 20 mL of water daily for 60 days, and then studied the brain tissue for gene chip analysis. [Results] Compared with the control group, the expressions of Thr, Dbh, Tph1, Drd3, Drd4, Adra1a, Adra2a, Adra2b and Adrb3 were down-regulated in the brain of mice infected with arsenite, while the expression of Htr1f, Htr4 and Htr7 was up-regulated. [Conclusion] As2O3 may result in the down-regulation of some neurotransmitter synthesis and catabolism enzymes in mouse brain tissue and interfere with the expression of some receptor genes.
