目的 :探讨癌胚抗原重组痘病毒 (carcinoembryonicantigenrecombinantvacciniavirus,CEA -rV)治疗CEA阳性肿瘤的作用部位及其CEA基因表达的情况。方法 :采用PCR法探测CEA rV的作用部位 ,RT -PCR检测接种CEA -rV鼠腹腔MΦ 内CEA的基因表达 ,免疫组化法检测各组小鼠腹腔MΦCEA的蛋白表达。结果 :腹腔接种的CEA -rV主要感染腹腔MΦ;皮下接种的CEA -rV主要感染皮肤局部组织。CEA -rV腹腔接种小鼠 ,其腹腔MΦ有CEAmRNA表达及CEA蛋白表达。结论 :推测无论通过腹腔接种还是通过皮肤局部接种 ,GEA -rV一般多在注射局部表达。CEA -rV不仅可以进入MΦ,而且其CEA基因可以有效地表达 ,并可使MΦ 表面分子MHC -Ⅱ与B7- 2表达增强 ,然后递呈CEA给Th淋巴细胞 ,使之分泌足够的IL - 2 ,激活Tc细胞分化为CTL ,杀伤肿瘤细胞
Objective : To investigate the role of carcinoembryonic antigen recombinantvacciniavirus (CEA-rV) in the treatment of CEA-positive tumors and their CEA gene expression. METHODS: The site of CEA rV was detected by PCR, and the gene expression of CEA in MΦ of CEA-rV mice was detected by RT-PCR. The expression of MΦCEA in the peritoneal cavity of mice was detected by immunohistochemistry. RESULTS: Intraperitoneal inoculation of CEA-rV was mainly associated with peritoneal MΦ; subcutaneously inoculated CEA-rV was predominantly infected with local tissue. CEA-rV was intraperitoneally inoculated in mice. The peritoneal MΦ had CEA mRNA expression and CEA protein expression. Conclusion : It is presumed that GEA-rV is usually localized in injections whether it is given by intraperitoneal injection or by local skin inoculation. CEA-rV can not only enter MΦ, but also its CEA gene can be efficiently expressed and enhance the expression of MΦ surface molecules MHC-II and B7-2, and then present CEA to Th lymphocytes, so that it can secrete enough IL-2. , Activate Tc cells to differentiate into CTL, kill tumor cells
