目的:研究3种五环三萜化合物齐墩果酸(OA)、乌苏酸(UA)、积雪草酸(AA)对肝细胞线粒体高通透性转运(MPT)的影响。方法:运用体外高钙诱发MPT模型,测定OA,UA,AA对肝细胞线粒体肿胀、线粒体膜电位耗散、线粒体内游离钙释放的影响。结果:正常小鼠肝细胞线粒体中加入50μmol.L-1 Ca2+即可引起明显的线粒体肿胀、膜电位耗散以及游离钙的释放,而50 mg.L-1 OA,UA,AA均可有效抑制上述过程,且3种化合物的抑制能力存在显著差异。结论:3种五环三萜化合物OA,UA,AA均具有显著对抗MPT,进而保护肝细胞线粒体的功能,且AA的作用强于UA及OA。
OBJECTIVE: To study the effects of oleanolic acid (OA), ursolic acid (UA) and asiatic acid (AA) of three pentacyclic triterpene compounds on mitochondrial permeability and transport of hepatocytes (MPT). Methods: The in vitro calcium-induced MPT model was used to determine the effects of OA, UA and AA on hepatocyte mitochondria swelling, mitochondrial membrane potential dissipation and mitochondrial free calcium release. Results: The addition of 50μmol.L-1 Ca2 + to mitochondria of normal mouse hepatocytes induced obvious swelling of mitochondria, dissipation of membrane potential and release of free calcium, while 50 mg.L-1 OA, UA and AA could be effectively inhibited The above process, and three kinds of compounds inhibitory ability there is a significant difference. Conclusion: The three pentacyclic triterpenoids OA, UA and AA all have significant antagonism against MPT, thus protecting the mitochondrial function of hepatocytes. AA is more effective than UA and OA.
