目的:观察参芪化瘀方对荷瘤小鼠肿瘤组织p53基因表达的影响,探讨参芪化瘀方对H22肝癌细胞作用的机制。方法:将H22肝癌荷瘤小鼠60只随机分为6组各10只:参芪化瘀方高剂量组、中剂量组、低剂量组,环磷酰胺注射用粉针剂(CTX)对照组,模型组,阴性对照组。参芪化瘀方高、中、低剂量组予以0.4mL中药(浓度分别为4.16g/mL、2.08g/mL、1.04g/mL)灌胃,CTX对照组予以0.2mL[小鼠给药剂量为20mg/(kg.d)]腹腔注射,模型组予以0.4mL生理盐水灌胃,每天1次,给药14天。免疫组织化学法检测肿瘤组织p53基因的表达,并观察其对脾脏指数、胸腺指数及肿瘤抑制率。结果:高、中、低剂量参芪化瘀方都能降低H22肝癌荷瘤小鼠肿瘤组织中p53基因的表达水平,且能升高小鼠的脾脏指数、胸腺指数。结论:参芪化瘀方对H22肝癌荷瘤小鼠肿瘤组织中p53基因的表达具有一定抑制作用,且能保护小鼠的免疫器官。
Objective: To observe the effect of Shenqi Huayu Recipe on p53 gene expression in tumor-bearing mice and to explore the mechanism of Shenqi Huayu Recipe on H22 hepatoma cells. Methods: Totally 60 H22 hepatoma tumor-bearing mice were randomly divided into 6 groups with 10 rats in each group: high dose of Shenqi Huayu Fang group, middle dose group, low dose group, CTX injection injection group, Model group, negative control group. Shenqi Huayu Fang high, medium and low dose group to 0.4mL Chinese medicine (concentration were 4.16g / mL, 2.08g / mL, 1.04g / mL) gavage, CTX control group was given 0.2mL [mice dosing 20mg / (kg.d)] intraperitoneal injection, the model group was given 0.4mL saline, once a day for 14 days. The expression of p53 gene in tumor tissue was detected by immunohistochemistry, and the spleen index, thymus index and tumor inhibition rate were observed. Results: High, medium and low dose of Shenqi Huayu Recipe could reduce the expression of p53 gene in tumor tissue of H22 hepatoma tumor-bearing mice and increase the spleen index and thymus index of mice. Conclusion: Shenqi Huayu Recipe can inhibit the expression of p53 gene in H22 hepatoma tumor-bearing mice and protect the immune organs of mice.
