目的探讨柴芩承气汤对急性坏死性胰腺炎(ANP)犬肺巨噬细胞白细胞介素1β(IL-1β)表达的影响。方法将15只比格犬随机分成对照组、模型组及柴芩承气汤组各5只,模型组及柴芩承气汤组采用50 mg/kg牛磺胆酸钠逆行胆管注射制作ANP模型。造模成功后,对照组及模型组经胃管注入生理盐水10 ml/kg,柴芩承气汤组经胃管注入柴芩承气汤10 ml/kg,各组均2 h注入1次,共3次。第6 h取各组犬血清检测IL-1β水平,并取肺组织分离巨噬细胞检测IL-1βmRNA及蛋白表达。结果模型组、柴芩承气汤组肺组织巨噬细胞IL-1βmRNA表达水平均较对照组明显下调(P<0.05);模型组肺组织巨噬细胞IL-1β蛋白表达及血清IL-1β水平显著高于对照组,柴芩承气汤组IL-1β蛋白表达及血清IL-1β水平显著低于模型组(P<0.05)。结论柴芩承气汤可下调ANP犬肺组织巨噬细胞IL-1β蛋白表达,降低血清IL-1β水平。
Objective To investigate the effect of Chai Qin Cheng Qi Decoction on the expression of interleukin-1β (IL-1β) in macrophages of acute necrotizing pancreatitis (ANP) in dogs. Methods Fifty Beagle dogs were randomly divided into control group, model group and Chaiqin Chengqi Decoction group. The model group and Chaiqin Chengqi Decoction group were treated with 50 mg / kg sodium taurocholate retrograde bile duct injection to make ANP model . After successful modeling, the control group and the model group were injected with 10 ml / kg normal saline via the gastric tube. The Chaiqin Chengqi Decoction group was injected with Chaiqin Chengqi Decoction 10 ml / kg through the gastric tube, A total of 3 times. The levels of IL-1β in serum of each group were measured at the 6th hour, and the macrophages were removed from the lung tissue to detect the expression of IL-1βmRNA and protein. Results The levels of IL-1βmRNA in lung tissue macrophages in model group and Chaiqin Chengqi Decoction group were significantly lower than those in control group (P <0.05). The expression of IL-1β in macrophage and the level of IL-1β The level of IL-1β and the level of IL-1β in Chaiqin Chengqi Decoction group were significantly lower than those in model group (P <0.05). Conclusion Chaiqin Chengqi decoction can down-regulate the expression of IL-1β in the lung tissue macrophages of ANP dogs and decrease the level of serum IL-1β.
