作者用豚鼠实验观察了抗原直接作用于内淋巴囊使内耳抗体水平升高。采用钥孔(虫戚)血蓝蛋白(keyhole limpet hemocyaninc KLH)抗原,同时用牛血清蛋白(BSA)作皮内免疫,抗体检测用酶联免疫吸附试验。动物分别给予500μgKLH/完全福氏佐剂和500μgBSA/完全福氏佐剂,直到能检测出循环中高抗KLH水平为止,BSA抗原敏化作为血清抗体的标记。之后,动物麻醉,显微镜下解剖出右侧内淋巴囊,用Hamilton注射器玻璃微管注入125μg/5μlKLH抗原,两周后分别从左、右耳取外淋巴和血清,对照动物是用PBS代替KLH。另外,手术堵塞右耳内淋巴管2周后,再
The authors used guinea pig experiments observed antigen direct action on the endolymphatic sac to raise the level of the inner ear antibody. Keyhole limpet hemocyaninc KLH antigen was used, while bovine serum albumin (BSA) was used for intradermal immunization. Antibody test was performed by enzyme linked immunosorbent assay. The animals were given 500 [mu] g KLH / complete Freund’s adjuvant and 500 [mu] g BSA / complete Freund’s adjuvant until the high circulating anti-KLH level was detectable, and BSA antigen sensitization was used as a marker for serum antibody. Animals were then anesthetized, the right endolymphatic sac was dissected microscopically, 125 [mu] g / 5 [mu] l KLH antigen was injected via Hamilton syringe glass microtubule, and two weeks later the perilymph and serum were removed from the left and right ears, respectively.
