建立高效液相色谱(HPLC)测定柯里拉京含量的方法,比较不同植物的柯里拉京含量,筛选出柯里拉京含量高的植物.并初步研究其抑制胃癌细胞增殖的活性.HPLC结果显示:龙眼核、叶下珠、龙眼壳、榄仁树和纤梗叶下珠的柯里拉京含量较高,依次为643,619,569,271和159μg/g.通过体外实验初步研究柯里拉京抑制胃癌细胞生长的活性,噻唑蓝(MTT)法测得柯里拉京对胃癌细胞SGC7901的半抑制浓度(IC50)值为(42.46±3.31)μmol/L;吖啶橙/溴化乙锭(AO/EB)和Hoechst 33258染色结果表明柯里拉京能诱导胃癌细胞SGC7901凋亡.摸索的HPLC条件能较快速、准确地检测出不同植物的柯里拉京含量,所测的12种材料中龙眼核的柯里拉京含量最高;柯里拉京具有抑制胃癌细胞SGC7901增殖的活性,并且能够诱导其凋亡.
To establish a HPLC method for the determination of corilagin content and compare the content of corilagin in different plants and screen the plants with high content of corilagin and to study the inhibitory effect on the proliferation of gastric cancer cells.HPLC results showed that: The content of corilagin in longan kernel, hyphae, longan shell, terminalis and stalk was higher, followed by 643, 619, 569, 271 and 159 μg / g respectively.Cellilagin inhibited the growth of gastric cancer cells by in vitro experiments, The half-inhibitory concentration (IC50) of corilagin to gastric cancer cell SGC7901 was (42.46 ± 3.31) μmol / L as measured by MTT assay. The results of acridine orange / ethidium bromide (EB) and Hoechst 33258 staining The results showed that corilagin could induce the apoptosis of SGC7901 cells in gastric cancer cells.It was found that the content of corilagin in different plants could be detected quickly and accurately by HPLC conditions. Rilking has the activity of inhibiting the proliferation of gastric cancer cell SGC7901 and inducing apoptosis.
