鸡胚神经管缺陷模型的建立

目的鸡胚胎内注射氨甲喋呤 (MTX) ,制作鸡胚神经管缺陷动物模型。 方法选用上海农业科学院“菜杭”鸡的受精蛋 5 5 2枚 ,分成 3组 ,分别为药物 (MTX)注射组、生理盐水注射对照组及正常对照组。药物注射时间选择在鸡胚孵化第 2~ 7d ,MTX剂量分别为 0 .0 1、0 .0 2、0 .0 5、0 .1及 0 .2 5mg/kg。孵化 3周后观察结果。  结果孵育第 3~ 6d注药的鸡 (剂量为 0 .0 1、0 .0 2、0 .0 5及 0 .1mg/kg)有神经管缺陷畸形产生 ,为脑膜脑膨出、骶尾骨退化等。 结论MTX能够制作出神经管缺陷鸡胚模型。 Objective To inoculate methotrexate (MTX) into chick embryo to make animal model of neural tube defects in chick embryo. Methods Five 5 2 fertilized eggs from “Cai Hang” chicken of Shanghai Academy of Agricultural Sciences were selected and divided into three groups: drug (MTX) injection group, saline injection control group and normal control group. Drug injection time selection in chicken embryo incubation 2 ~ 7d, MTX doses were 0. 0 1,0. 0 2,0. 0 5,0 .1 and 0. 25mg / kg. After 3 weeks of incubation, the results were observed. Results The chickens (doses 0. 0 1. 0 2 .0 0. 0 and 0 1 mg / kg) that were injected on the 3rd to 6th day had neural tube defects and malformations, which were meningoencephalocele and sacrococcygeal degeneration Wait. Conclusion MTX can produce neural tube defects chicken model.
