目的:研究亮菌多糖ATPSⅡ-2的抗肿瘤活性及其对荷瘤小鼠免疫功能的影响。方法:以4种人肿瘤细胞株为模型,采用噻唑蓝法检测体外抗肿瘤活性;建立小鼠S180荷瘤模型,以抑瘤率检测ATPSⅡ-2的体内抗肿瘤作用,以对荷瘤小鼠脾脏指数和胸腺指数的影响评价ATPSⅡ-2对小鼠免疫功能的作用;ELISA法检测对荷瘤小鼠血清中肿瘤坏死因子-α(TNF-α)和白细胞介素-2(IL-2)水平的影响。结果:ATPSⅡ-2对人肺癌细胞株的生长具有抑制作用,对Hela,K562和Hep G2细胞的增殖无明显的抑制作用;ATPSⅡ-2高、中、低剂量组对S180的抑瘤率分别为52.7%,45.9%和12.8%,且能提高荷瘤小鼠脾脏和胸腺指数。与阴性组比较,ATPSⅡ-2多糖高、中剂量组可提高荷瘤小鼠血清中细胞因子IL-2和TNF-α的水平,差异有统计学意义。结论:亮菌多糖ATPSⅡ-2有一定的细胞毒活性,有较强的体内抗肿瘤活性,其机制与调节血清细胞因子水平、增强机体免疫功能有关。
Objective: To study the anti-tumor activity of Polysaccharide ATPSⅡ-2 and its effect on immune function of tumor-bearing mice. METHODS: Four human tumor cell lines were used as the model and the antitumor activity in vitro was detected by the method of thiazolyl blue. The S180 tumor-bearing mice model was established and the anti-tumor effect of ATPSⅡ-2 was detected by inhibition rate. Spleen index and thymus index to evaluate the effect of ATPSⅡ-2 on immune function in mice; ELISA assay was used to detect the levels of tumor necrosis factor-α (TNF-α) and interleukin-2 (IL-2) Horizontal impact. RESULTS: ATPSⅡ-2 had inhibitory effect on the growth of human lung cancer cell lines and had no significant inhibitory effect on the proliferation of Hela, K562 and Hep G2 cells. The inhibitory rates of ATPSⅡ-2 to S180 were 52.7%, 45.9% and 12.8%, respectively, and could increase the spleen and thymus index of tumor-bearing mice. Compared with the negative group, high and medium dose of ATPSⅡ-2 polysaccharide could increase the level of cytokines IL-2 and TNF-α in tumor-bearing mice, the difference was statistically significant. CONCLUSION: The polysaccharide ATPSⅡ-2 has some cytotoxic activity and strong anti-tumor activity in vivo. The mechanism of ATPSⅡ-2 is related to the regulation of serum cytokines and the enhancement of immune function.
