本文以锦绣杜鹃花为原料,以游离氨基酸提取率为衡量指标,选取超声时间、液料比、超声温度、乙酸浓度为考察因素,在单因素试验基础上运用L9(34)正交试验设计,确定杜鹃花游离氨基酸测定过程的最佳提取工艺为:超声温度为60℃,料液比为1:40,超声时间为30min,提取温度为60℃,乙酸溶度为5%。应用正交试验优化工艺对游离氨基酸的提取进行验证,取得了较好的效果,表明此优化提取工艺稳定、可行。
Taking Splendid Rhododendron as raw material and free amino acid extraction rate as the measurement index, the ultrasonic time, the ratio of liquid to material, the temperature of ultrasound and the concentration of acetic acid were selected as the investigation factors. Based on the single factor experiment, the orthogonal design of L9 (34) The optimal extraction process of determination of free amino acids in rhododendron was determined as follows: the ultrasonic temperature was 60 ℃, the solid-liquid ratio was 1:40, the ultrasonic time was 30 min, the extraction temperature was 60 ℃ and the acetic acid solubility was 5%. The application of orthogonal test optimization process to verify the extraction of free amino acids, and achieved good results, indicating that the optimal extraction process is stable and feasible.
