目的:建立肿瘤坏死因子(TNF-α)损伤心脏微血管内皮细胞(CMEC)模型的方法学,为体外筛选具有血管保护作用的药物提供科学的实验载体与平台。方法:采用CMEC为研究对象,以MTT法检测细胞活力,观察200、400、800U/ml TNF-α对CMEC活力及乳酸脱氢酶(LDH)的影响。结果:TNF-α能够损伤CMEC,降低CMEC的活力,促进LDH的释放,以TNF-α400U/ml浓度组损伤最明显。结论:根据TNF-α对CMEC作用的量效关系,选择400U/ml为最合适的细胞刺激条件。
OBJECTIVE: To establish a methodology for detecting cardiac microvascular endothelial cells (CMECs) damaged by tumor necrosis factor (TNF-α) and provide a scientific experimental carrier and platform for screening drugs with vascular protection in vitro. Methods: CMEC was used as the research object and the cell viability was detected by MTT assay. The effects of 200,400,800 U / ml TNF-α on the activity of CMEC and lactate dehydrogenase (LDH) were observed. Results: TNF-α could damage CMEC, reduce the activity of CMEC and promote the release of LDH. The most obvious damage was at the concentration of TNF-α400U / ml. Conclusion: According to the dose-effect relationship of TNF-α on CMEC, 400U / ml is the most suitable cell stimulation condition.
