目的探讨血管紧张素Ⅱ(AngⅡ)在不同浓度和不同作用时间下对血管内皮细胞凋亡及凋亡调控基因的作用及依诺肝素对其影响。方法采用流式细胞技术测定了在AngⅡ不同浓度和不同作用时间下血管内皮细胞凋亡率和Fas和Bcl2表达的变化及依诺肝素对其的影响。结果AngⅡ可明显促进血管内皮细胞凋亡,当AngⅡ终浓度为10-8、10-7、10-6、10-5及10-4时,内皮细胞凋亡分别为186±018、3062±420、167±015、3008±441,Fas(113±016、1485±147、113±016、1567±197)和Bcl2(238±043、962±181、238±043、1233±173)的表达剂量依赖性和时间依赖性;依诺肝素对AngⅡ的有抑制作用(3062±420,1831±264;1485±147,728±136;962±181,1605±179)。结论血管紧张素II具有明显的促进血管内皮细胞凋亡和凋亡调控基因表达的作用;依诺肝素有一定抑制内皮细胞凋亡和凋亡调控基因表达的作用。
Objective To investigate the effects of angiotensin Ⅱ (Ang Ⅱ) on apoptosis and apoptosis-regulating genes of vascular endothelial cells (ECs) under different concentrations and different time periods and the effect of enoxaparin on them. Methods Flow cytometry was used to determine the changes of apoptosis rate and expression of Fas and Bcl-2 in endothelial cells under different concentrations of AngⅡ and the effect of enoxaparin. Results AngⅡ could significantly promote the apoptosis of vascular endothelial cells. When the final concentration of AngⅡ was 10-8, 10-7, 10-6, 10-5 and 10-4, the apoptosis of endothelial cells were 186 ± 018, 3062 ± 420 , 167 ± 015, 3008 ± 441, Fas (113 ± 016, 1485 ± 147, 113 ± 016, 1567 ± 197) and Bcl2 (238 ± 043, 962 ± 181, 238 ± 043, 1233 ± 173) (3062 ± 420,1831 ± 264; 1485 ± 147,728 ± 136; 962 ± 181,1605 ± 179) of AngⅡ. Conclusion Angiotensin II can significantly promote the apoptosis of vascular endothelial cells and the expression of apoptosis-controlling genes. Enoxaparin can inhibit the apoptosis of endothelial cells and regulate the gene expression of apoptosis.
