目的探讨胆固醇缺乏时对Jurkat细胞增殖功能及凋亡的影响。方法用Lovastatin抑制Jurkat细胞内源性胆固醇的合成 ,以低密度脂蛋白受体(LDL R)介导的低密度脂蛋白(LDL)内吞提供细胞外源性胆固醇 ,细胞处理1~3d后 ,用流式细胞仪分析各细胞周期相细胞分布 ,并定量分析胆固醇缺乏对处理3d后细胞凋亡的影响。结果Jurkat经去脂血清培养基加Lovastatin处理3d后 ,细胞受阻于G0/G1 期 ,使细胞增殖明显受抑 ,且诱导细胞凋亡的出现 ,加入LDL可以部分逆转上述变化。结论无论是内源性还是外源性胆固醇的缺乏都能抑制Jurkat细胞的增殖 ,并能在一定程度上诱导细胞凋亡的发生 ,且这一作用与细胞胆固醇的缺乏程度呈正性相关。
Objective To investigate the effects of cholesterol deficiency on proliferation and apoptosis of Jurkat cells. Methods Lovastatin inhibited the synthesis of endogenous cholesterol in Jurkat cells and provided LDL-mediated endocytosis of low density lipoprotein (LDL) to provide extracellular cholesterol. After 1 to 3 days of cell processing, The cell cycle distribution of cells was analyzed by flow cytometry, and the effect of cholesterol deficiency on the apoptosis of cells after 3-day treatment was quantitatively analyzed. Results After treated with Lovastatin for 3 days, Jurkat cells were blocked in G0 / G1 phase, inhibited cell proliferation and induced apoptosis. Adding LDL partially reversed these changes. Conclusion Both endogenous and exogenous cholesterol deficiency can inhibit the proliferation of Jurkat cells and induce apoptosis to a certain extent, and this effect is positively correlated with the lack of cellular cholesterol.
