目的:探讨颈髓损伤后颈髓线粒体膜流动性变化与线粒体呼吸功能的关系。方法:采用Allen法造成猫颈髓损伤,观察颈髓伤后线粒体膜流动性、磷脂酶A2(PLA2)活性、线粒体呼吸控制率(RCR)、磷氧比值(P/O)、氧化磷酸化效率(OPR)的变化。结果:颈髓损伤后2~72h线粒体膜PLA2活性较假手术对照组明显增高(P<005),而线粒体膜流动性、线粒体呼吸控制率(RCR)、磷氧比值(P/O)、氧化磷酸化效率(OPR)均较假手术对照组明显降低(P<005)。结论:表明颈髓损伤后PLA2激活与膜流动性降低密切相关,提示颈髓线粒体膜流动性降低在颈髓损伤后线粒体呼吸功能损害的病理生理过程中起重要作用。
Objective: To investigate the relationship between mitochondrial membrane fluidity changes and mitochondrial respiratory function after cervical spinal cord injury. Methods: The cervical spinal cord injury was induced by Allen method. The mitochondrial membrane fluidity, phospholipase A2 (PLA2) activity, mitochondrial respiratory control rate (RCR), P / O ratio and oxidative phosphorylation efficiency (OPR) changes. RESULTS: PLA2 activity in mitochondrial membrane between 2 and 72 h after SCI was significantly higher than that in sham operation group (P <0.05), while mitochondrial membrane fluidity, mitochondrial respiratory control rate (RCR), P / O ratio, Phosphorylation efficiency (OPR) were significantly lower than the sham control group (P <005). CONCLUSIONS: PLA2 activation is associated with decreased membrane fluidity after cervical spinal cord injury, suggesting that reduced fluidity of the cervical spinal cord mitochondria may play an important role in the pathophysiology of mitochondrial respiratory dysfunction after cervical spinal cord injury.
