目的 研究正常肝细胞和肝癌细胞流变特性与细胞骨架结构的相关性。方法 以微管吸吮技术测定两种细胞的粘弹性系数和细胞在2μg/ml Ⅳ型胶原则1.25 μg/ml层粘连蛋白(laminin, LN)复台裱衬表面上的粘附力,以秋水仙素(colchicine,Col)和细胞松弛素D(cytochlasin,CD)处理细胞研究其对细胞粘弹性系数和粘附力的影响。结果 在质量浓度为1~60 mg/L的Col作用下,肝细胞弹性系数及粘附力总体上明显增高或趋于增高,而肝癌细胞粘弹性系数及粘附力明显降低;在质量浓度为0.25~ 500 mg/L的CD作用下,两种细胞粘弹性系数及粘附力均明显降低,肝癌细胞弹性系数K1、K2值及粘附力的下降幅度明显大于肝细胞相应值的下降幅度。细胞在Ⅳ型胶原/LN复合裱衬表面的粘附力与细胞粘弹性系数呈相同的变化趋势,肝癌细胞粘附力变化与细胞弹性系数K1、 K2的变化呈高度正相关(P<0.01)。结论 在CD和Col作用下肝细胞和肝癌细胞的流变特性呈不同的效应方式或强度,反映了肝细胞和肝癌细胞细胞骨架结构和机能状态的差异。
Objective To study the correlation between rheological characteristics and cytoskeletal structure in normal liver cells and hepatoma cells. Methods The viscoelastic coefficient of two cells was determined by micropipette sucking technique and the adhesion of cells to 1.25 μg/ml laminin (LN) reconstituted tarpaulin lining surface at 2 μg/ml IV adhesive principle. The effects of colchicine (Col) and cytochlasin (CD) on the viscoelastic coefficient and adhesion of cells were studied. Results Under the action of Col with a concentration of 1~60 mg/L, the elasticity coefficient and adhesion of hepatocytes were significantly increased or tended to increase, while the viscoelastic coefficient and adhesion of hepatoma cells were significantly decreased. Under the action of 0.25 to 500 mg/L of CD, the viscoelastic coefficient and adhesion of both cells were significantly reduced. The decrease of the elasticity coefficients K1 and K2 and the adhesion of hepatoma cells was significantly greater than that of the corresponding values of hepatocytes. Magnitude. The adhesion of cells on the surface of type IV collagen/LN composite lining showed the same tendency with the cell viscoelastic coefficient, and the change of adhesion of hepatoma cells was highly positively correlated with the changes of cell elasticity coefficients K1 and K2 (P<0. 01). Conclusion The rheological characteristics of hepatocytes and hepatoma cells under the action of CD and Col showed different effects or intensity, reflecting the differences in the cytoskeletal structure and function of hepatocytes and hepatoma cells.
