目的:探讨HT-29肠癌细胞、正常肠上皮细胞及ECV-304血管内皮细胞中E-selectin、Integrinβ1及ICAM-1的表达状态。方法:采用Nothern Blotting方法检测HT-29肠癌细胞、正常肠上皮细胞和ECV-304血管内皮细胞中E-selectin、Integrinβ1及ICAM-1mRNA表达水平,采用ELISA法定量分析其表达含量。结果:HT-29肠癌细胞、正常肠上皮细胞和ECV-304血管内皮细胞均有E-selectin、Integrinβ1及ICAM-1基因表达。ELISA定量测定3个粘附分子表达水平,HT-29肠癌细胞均高于正常肠上皮细胞和ECV-304血管内皮细胞,分别存在显著性差异(P<0.05)。结论:E-selectin、Integrinβ1、ICAM-1可能与肿瘤细胞转移有关。
Objective: To investigate the expression of E-selectin, Integrinβ1 and ICAM-1 in HT-29 colorectal cancer cells, normal intestinal epithelial cells and ECV-304 vascular endothelial cells. Methods: The expression of E-selectin, Integrinβ1 and ICAM-1 mRNA in HT-29 colorectal cancer cells, normal intestinal epithelial cells and ECV-304 vascular endothelial cells were detected by Nothern Blotting. The expression levels of integrinβ1 and ICAM-1 were detected by ELISA. Results: The expression of E-selectin, Integrinβ1 and ICAM-1 were detected in HT-29 colon cancer cells, normal intestinal epithelial cells and ECV-304 vascular endothelial cells. ELISA showed that there were significant differences in the expression of three adhesion molecules between HT-29 colon cancer cells and normal intestinal epithelial cells and ECV-304 vascular endothelial cells (P <0.05). Conclusion: E-selectin, Integrinβ1 and ICAM-1 may be involved in tumor cell metastasis.
