C组肠道病毒(Human enterovirus C,HEV-C)共包括18个血清型,通过使用现有的组合血清进行中和试验的方法难以鉴定它们的型别。为探讨HEV-C山东地方株的基因型分布及分子进化特征,本研究对1994年~2008年急性弛缓性麻痹(Acute flaccid paralysis,AFP)监测系统中分离到的未定型的毒株,采用HEV-C VP1完整编码区的特异性引物,进行逆转录-聚合酶链反应(Reverse transcriptase-polymerase chain reaction,RT-PCR),产物进行序列测定后,通过NCBI服务器中BLAST程序进行型别鉴定,并与其他HEV-C各基因型代表株进行比较并构建系统发生树。共获得12株HEV-C山东地方株,分别属于4个基因型:CA20、CA21,CA24和EV96。各基因型内山东地方分离株跟原型株相比基因序列同源性较低,系统发生学分析显示HEV-C山东地方株各血清型与原型株亲缘关系较远,提示山东省存在不同基因特征的HEV-C传播。
Group C enterovirus (HEV-C) includes a total of 18 serotypes. It is difficult to identify their genotype by the neutralization test using the existing combination sera. In order to investigate the genotypic distribution and molecular evolution of HEV-C Shandong local strains, we studied the genotypic distribution and molecular evolution of HEV-C Shandong local strains. The untreated strains isolated from AFP in AFP from 1994 to 2008 were cultured in HEV-C -C VP1 complete coding region of specific primers, reverse transcriptase-polymerase chain reaction (Reverse transcriptase-polymerase chain reaction, RT-PCR), the product sequence determination, by NCBI server BLAST program type identification, and The other HEV-C genotypes were compared with each other and the phylogenetic tree was constructed. Twelve HEV-C Shandong local strains were obtained, belonging to four genotypes: CA20, CA21, CA24 and EV96 respectively. Shandong province isolates in each genotype had lower homology with the original strains. Phylogenetic analysis showed that the serotypes of HEV-C Shandong local strains were distantly related to the prototype strains, suggesting that there are different genetic characteristics in Shandong Province HEV-C transmission.
