利用RAPD技术对松材线虫(Bursaphelenchusxylophilus)和拟松材线虫(B.mucrona-tus)的分离物基因组DNA进行了DNA片段的随机扩增,以确定种间及种内群体的亲缘关系和筛选具有种鉴定特征的分子标记.通过对40个10聚体随机引物的筛选,应用8个引物对两种线虫种间及种内分离物扩增片段的遗传分析显示:两种间群体RAPD扩增片段的共享度为40%~56%,而在松材线虫的不同分离物之间的共享度为60%~84%;其中有一个引物(P253)可在松材线虫和拟松材线虫种间产生具有种鉴定特征的特异性RAPD片段,在松材线虫的不同分离物中均可产生一约700bp的特异性片段,而在拟松材线虫中产生了两个分别约为1300bp和1900bp的特异性片段.
The RAPD technique was used to randomly amplify DNA fragments of genomic DNA isolated from Bursaphelenchus xylophilus and B. mucrona-tus to determine the phylogenetic relationships and screening of Molecular identification of species. Genetic analysis of amplified fragments of two species of interspecific and intraspecific isolates using 8 primers by screening 40 10-mer random primers showed that the share of RAPD fragments between the two species was 40% 56%, while the share of different isolates of Bursaphelenchus xylophilus is 60% -84%. One of the primers (P253) can be used to produce a species-specific characteristic between Bursaphelenchus xylophilus Sex RAPD fragments produced a specific fragment of about 700 bp in different isolates of B. xylophilus and two specific fragments of approximately 1300 bp and 1900 bp in B. subtilis, respectively.
