目的 :探讨链霉素耳中毒豚鼠耳蜗外毛细胞外向钾通道的变化。方法 :应用听觉脑干诱发电位和全细胞膜片钳制技术。结果 :链霉素耳中毒豚鼠体重明显降低 ;在以 2kHz为主的click刺激时听阈明显升高 ;从链霉素耳中毒豚鼠中分离的外毛细胞数明显减少 ,且以直径较长细胞为主 ;链霉素显著降低豚鼠耳蜗外毛细胞延迟外向K+ 电流 ;链霉素耳中毒豚鼠耳蜗外毛细胞Ca2 + 敏感外向K+ 通道和延迟外向K+ 通道的反转电位与正常对照组相比没有差异。结论 :链霉素对外毛细胞外向钾通道的阻断作用是其耳毒性的基础 ,但不是细胞致死的直接原因。
Objective: To investigate the changes of extracellular potassium channels in cochlear outer hair cells of streptomycin ototoxicity guinea pigs. Methods: Auditory brainstem response and whole-cell patch-clamp technique were used. Results: The body weight of streptozotocin-induced guinea pigs was significantly lower than that of the control group. The threshold was significantly increased at the frequency of 2 kHz. The number of outer hair cells isolated from streptomycin ototoxicity guinea pigs was significantly decreased. Main; streptomycin significantly reduced outward outward cochlear outer hair cells in guinea pigs outward K + current; streptomycin ototoxicity guinea pig cochlear outer hair cells Ca2 + sensitive outward K + channels and delayed outward K + channel reversal potential compared with the normal control group no difference . Conclusion: The blocking effect of streptomycin on the outward potassium channel of the outer hair cell is the basis of its ototoxicity, but it is not the direct cause of cell death.
