目的优化痛安注射液组方药材白屈菜提取物精制纯化工艺,提高白屈菜提取物中白屈菜碱的量。方法分别考察平板直联式离心、高速管式离心、碟片离心不同离心技术方法,对白屈菜酸沉液分离的效果;分别考察不同pH值酸水液对白屈菜萃取干膏溶解效果的影响;分别考察减压滤过、高速管式离心、平板直联式离心不同分离方式对白屈菜酸水溶解液分离的效果;以白屈菜碱转移率、干膏得率为评价指标,确定可行的白屈菜碱精制纯化工艺。结果采用碟片离心技术对白屈菜酸沉液分离效果最好;酸水液pH值在3~4时酸溶解效果及白屈菜碱转移率最佳;高速管式离心对白屈菜酸水溶解液分离效果最好。结论该工艺制备白屈菜提取物,操作简单,稳定可行。
OBJECTIVE To optimize the purification and purification process of chelerythrine extract of Tong’an injection and to increase the content of chelidon in chelerythrine extract. Methods The effects of different concentrations of sour water solution on the dissolution of Celastrus glauca extract were investigated respectively by direct centrifugal plate centrifugation, high-speed tube centrifugation and eccentric centrifugation by disc centrifugation. The effect of different separation methods of chelerythrine acid solution on vacuum separation filtration, high-speed tube centrifugation and flat-bed direct-coupled centrifugation was investigated respectively. The chelerythrine transfer rate and the dry extract yield were evaluated to determine the feasibility Chelidon purification process. Results The disc centrifuge technique was the best for the separation of celandine acid precipitation solution. The optimum acid-dissolving effect and chelidonine transfer rate were obtained when pH value of acid-water solution was 3 ~ 4; Liquid separation best. Conclusion The process for the preparation of celandine extract, simple, stable and feasible.
