利用陈永强“氯仿—异戊醇—核酸酶法”首次对普通小麦、燕麦、水稻、玉米、冰草5种作物以及同种作物的不同品种和不同器官的DNA进行提取,结果表明,不同作物的幼穗DNA浓度不同,冰草幼穗DNA浓度(0.75μg/ml)是燕麦幼穗的4倍,玉米和水稻的幼穗DNA浓度很接近,为0.363μg/ml和0.378μg/ml。同一种作物不同品种的幼穗DNA浓度也不同,爆裂玉米品种幼穗DNA浓度(0.55μg/ml)是掖单19号的3.1倍,水稻品种216的DNA浓度高于宁糯4号和51,通北燕麦的幼穗DNA浓度高于其它3种燕麦。同一种作物不同器官的DNA浓度也不一致,幼穗的DNA浓度高于幼茎。
Using the method of “chloroform-isoamyl-nuclease” by Chen Yongqiang, we first extracted the DNA from different varieties and organs of five kinds of crops such as common wheat, oat, rice, corn and grass and the same crops. The results showed that different crops DNA concentrations of young spikelets were different, and the DNA concentration of young spikelets (0.75μg / ml) was 4 times that of oats, and the young spike DNA concentrations of maize and rice were close to 0.363μg / ml and 0.378μg / ml . The DNA concentration of young spike of different breeds of the same crop was also different. The DNA concentration of young spike (0.55μg / ml) was 3.1 times higher than that of Yedan 19 and the DNA concentration of 216 was higher than Ningnuo 4 And 51, the DNA concentration of young ears in Tongbei oats was higher than that of the other three kinds of oats. The same crop of different organs of the DNA concentration is inconsistent, young ears of DNA concentration is higher than young stems.
