目的探讨糖尿病肾病大鼠模型肾小管间质纤维化与瘀血阻络的相关性。方法 30只SD大鼠随机平分为三组-瘀血阻络型组(A组)、非瘀血阻络型组(B组)、正常组(C组),均予高糖高脂饲料喂养,A组与B组采用一次性腹腔注射链脲佐菌素(STZ)诱导建立糖尿病肾病模型,观察大鼠行为,分别测定随机血糖、24h尿微量白蛋白含量;取大鼠的肾皮质,病理切片染色观察肾小管间质的变化;也采用western blot检测各组大鼠肾组织TGF-β1的表达情况。结果 A组与B组的不同时间点的血糖明显高于C组(P<0.05),A组与B组的组间对比无明显差异(P>0.05)。A组与B组的24h尿微量白蛋白定量分别为(24.20±4.10)mg/24h和(24.29±3.89)mg/24h,都明显高于C组的(5.93±2.14)mg/24h(P<0.05)。A组与B组的TGF-β1表达量明显高于C组(P<0.05),同时A组的TGF-β1表达量也高于B组(P<0.05)。形态学观察显示相对于B组,A组的肾小球体积增大,基底膜增厚,系膜基质增多。结论糖尿病肾病大鼠模型中瘀血阻络型的TGF-β1表达量相对比较高,伴随有肾小管间质纤维化,可导致大鼠病情恶化,也确认了糖尿病肾病瘀血阻络型的存在。
Objective To investigate the correlation between tubulointerstitial fibrosis and blood stasis resistance in diabetic nephropathy rat model. Methods Thirty SD rats were randomly divided into three groups: blood stasis group (group A), non-blood stasis group (group B) and normal group (group C) , Group A and group B were induced by streptozotocin (STZ) intraperitoneally once a day to establish diabetic nephropathy model. The behavior of rats was observed and the contents of random blood glucose and 24h urinary albumin were measured respectively. The renal cortex, pathology Slides were stained to observe the changes of tubulointerstitial. Western blot was also used to detect the expression of TGF-β1 in renal tissues. Results The blood glucose in group A and group B at different time points was significantly higher than that in group C (P <0.05). There was no significant difference between group A and group B (P> 0.05). The urinary microalbuminuria of group A and group B were (24.20 ± 4.10) mg / 24h and (24.29 ± 3.89) mg / 24h respectively, which were significantly higher than that of group C (5.93 ± 2.14) mg / 24h (P < 0.05). The expression of TGF-β1 in group A and group B was significantly higher than that in group C (P <0.05), while the expression of TGF-β1 in group A was also higher than that in group B (P <0.05). Morphological observation showed that compared with group B, glomerular volume of group A increased, basement membrane thickening and mesangial matrix increased. Conclusion The expression of TGF-β1 in blood stasis syndrome in rats with diabetic nephropathy is relatively high. With the development of tubulointerstitial fibrosis, it may lead to the worsening of the disease in rats and the existence of the obstruction-occlusion pattern of diabetic nephropathy .
