作者探讨了干细胞因子(SCF)单独及与IL3、IL6和GM-CSF结合对LT12白血病造血的体外调控效应。结果发现,SCF可直接抑制LT12细胞的CFU-L集落形成、DNA合成及增殖动力学。100ng/mlSCF导致实质性抑制效应,尽管这种抑制活性低于IL3、IL6及GM-CSF。此外,SCF可增强这三种因子的抑制活性,与IL6或GM-CSF联合应用所引起的抑制效应,要高于与IL3结合;SCF与两个以上因子合用所产生的抑制更加显著。结合作者先前的研究结果认为,在BNML大鼠白血病造血中,SCF具有两种截然不同的生物效应:有效地刺激正常造血又可抑制白血病造血。
The authors investigated the in vitro regulatory effect of stem cell factor (SCF) on the hematopoiesis of LT12 leukemia alone and in combination with IL3, IL6 and GM-CSF. The results showed that SCF could directly inhibit CFU-L colony formation, DNA synthesis and proliferation kinetics of LT12 cells. 100 ng / ml SCF resulted in a substantial inhibitory effect, although this inhibitory activity was lower than that of IL3, IL6 and GM-CSF. In addition, SCF can enhance the inhibitory activity of these three factors, with IL6 or GM-CSF combination of the inhibition caused by the effect is higher than with IL3; SCF combination of two or more factors produced more significant inhibition. In combination with the authors’ previous findings, SCF has two distinct biological effects on hematopoietic leukemia in BNML rats: effective stimulation of normal hematopoiesis and inhibition of leukemia hematopoiesis.
