目的调查四川地区宫颈癌患者组织中高危人乳头瘤病毒(HPV)33、52和58亚型的感染状况及E6基因突变特性,为制备适合中国人群的HPV疫苗提供流行病学资料。方法采用PCR技术,并结合基因测序分析。结果对四川地区2003~2004年60份宫颈癌患者癌组织DNA进行HPV检测,获得HPV52和HPV58亚型各4份,检出率为6·7%;HPV33亚型1份,检出率为1·7%。对所获得的各份HPV33、52和58E6基因测序,均有碱基突变。结论HPV52型感染率在四川地区相对较高。与GenBank标准株相比,四川地区发现的HPV33和HPV52E6基因有多处突变,与日本发现的HPV33和HPV52突变株相近。
Objective To investigate the infection status and mutation characteristics of E6 gene in high risk human papillomavirus (HPV) types 33, 52 and 58 in patients with cervical cancer in Sichuan province and provide epidemiological data for preparing HPV vaccine suitable for Chinese population. Methods PCR technology, combined with gene sequencing analysis. Results The DNA of 60 cases of cervical cancer from 2003 to 2004 in Sichuan Province was tested for HPV. Four HPV52 and HPV58 subtypes were obtained, the detection rate was 6.7% and the HPV33 subtype was 1 · 7%. The obtained HPV33, 52 and 58E6 gene sequencing, there are base mutations. Conclusion HPV52 infection rate is relatively high in Sichuan. Compared with the GenBank standard strains, the HPV33 and HPV52E6 genes found in Sichuan have multiple mutations, similar to the HPV33 and HPV52 mutations found in Japan.
