观察耳蜗外毛细胞发生凋亡时听力改变情况。方法 :对 2 0只豚鼠药物造模 ,诱发耳蜗外毛细胞发生凋亡。应用TUNEL技术观察凋亡表达 ,测试ABR阈值观察听力变化。结果 :应用丁胺卡那霉素 1天即可诱发豚鼠耳蜗外毛细胞发生凋亡 ,连续应用 3d ,耳蜗外毛细胞凋亡呈强阳性表达 ,但ABR阈值无明显改变 ;随着用药时间延长 ,凋亡细胞数目增加 ,甚至出现部分毛细胞缺失现象 ,此时ABR阈值明显升高。结论 :耳蜗外毛细胞发生凋亡早期对豚鼠听力无明显影响 ,随着耳毒性药物应用时间延长 ,豚鼠ABR阈值升高可能存在两种原因 :毛细胞凋亡或毛细胞坏死。
To observe the changes of hearing in the occurrence of apoptosis of cochlear outer hair cells. Methods: Twenty guinea pigs were induced to induce apoptosis of outer hair cells. TUNEL technique was used to observe the expression of apoptosis, and the ABR threshold was tested to observe the change of hearing. Results: The apoptosis of guinea pig cochlear outer hair cells was induced by amikacin for one day. The apoptosis of cochlear outer hair cells was strongly positive for 3 consecutive days, but the ABR threshold did not change significantly. , The number of apoptotic cells increased, and even some of the hair loss cells, ABR threshold increased significantly. CONCLUSION: There is no obvious effect on the hearing of guinea pigs during the early stage of apoptosis of cochlear outer hair cells. There are two possible reasons for the increase of ABR threshold in guinea pig with the prolonged ototoxicity: hair cell apoptosis or hair cell necrosis.
