目的:观察龙首参葛丸对血管性痴呆大鼠认知障碍的改善作用。方法:改良2-VO法复制血管性痴呆模型大鼠,分为模型组、盐酸多奈哌齐片阳性对照组及龙首参葛丸高、中、低剂量组,另设假手术组。各组大鼠灌胃给药,1次/d,连续6 w。大鼠给药第36天进行Morris水迷宫实验,第36~41天进行定向航行实验及空间探索实验,及海马长时程升高(LTP)实验。以上实验结束后,检测各组大鼠血清及海马组织SOD、MDA、GSH、GSH-Px、NO及NOS水平。结果:龙首参葛丸可缩短血管性痴呆模型大鼠Morris水迷宫实验逃避潜伏期、延长第三象限活动时间、增加穿越平台次数,延长海马长时程升高实验10、30、60、90 min的PS幅值;同时升高血清和海马组织SOD、GSH及GSH-Px水平,降低血清和海马组织MDA、NO及NOS水平。结论:龙首参葛丸可改善血管性痴呆模型大鼠的认知障碍,提高其学习记忆能力和空间探索能力及机体清除自由基抗氧化能力。
Objective: To observe the effect of Longshensen Ge pill on cognitive impairment in vascular dementia rats. Methods: The model of vascular dementia was established by modified 2-VO method and divided into model group, donepezil hydrochloride positive control group, Long Shuan Shen Ge Wan high, medium and low dose groups, and another sham operation group. Rats in each group were given gavage once a day for 6 weeks. Morris water maze test was performed on the 36th day in rats, directional navigation experiments and space exploration experiments were performed on days 36 to 41, and hippocampal long-term elevated (LTP) experiments were performed. After the above experiment, the levels of SOD, MDA, GSH, GSH-Px, NO and NOS in serum and hippocampus were detected. Results: Longshen Geshu pill can shorten the vascular dementia model rats Morris water maze test escape latency, extend the third quadrant activity time, increase the number of crossing the platform to extend the hippocampal long-term high test 10,30,60,90 min PS, while increasing SOD, GSH and GSH-Px levels in serum and hippocampus, and decreasing MDA, NO and NOS in serum and hippocampus. Conclusion: Radix Ginseng Pill can improve cognitive impairment in rats with vascular dementia, enhance learning and memory abilities, space exploration ability and anti-oxidation ability of free radicals in the body.
