目的采用HPLC法测定人血液和尿液中的倍他米松。方法以乙酸乙酯为萃取剂、泼尼松为内标。用Welchrom C18柱,甲醇-水(60∶40)为流动相,检测波长240 nm。结果倍他米松在血液和尿液中的线性范围为0.1~20μg.mL-1(r2>0.999);检测限均为0.02μg.mL-1;平均方法回收率为91%~104%;日内、日间RSD≤4.6%;萃取回收率≥86.3%。结论所建方法准确、灵敏、简便,适用于血液、尿液中倍他米松的分析。
Objective To determine betamethasone in human blood and urine by HPLC. Methods Ethyl acetate as extractant and prednisone as internal standard. Using a Welchrom C18 column, methanol-water (60:40) as the mobile phase, the detection wavelength was 240 nm. Results The linear range of betamethasone in blood and urine was 0.1 ~ 20μg.mL-1 (r2> 0.999). The detection limits were 0.02μg.mL-1. The average recoveries were 91% -104% , Daytime RSD≤4.6%; extraction recovery≥86.3%. Conclusion The proposed method is accurate, sensitive and simple and suitable for the analysis of betamethasone in blood and urine.
