目的:研究血红素氧合酶1(HO-1)、血红素氧合酶2(HO-2)在雌性去势大鼠阴道平滑肌中的表达,探讨雌激素对阴道平滑肌HO-1、HO-2表达的影响。方法:雌性8周龄SD大鼠随机分为A(去卵巢2周组)、B(去卵巢4周组)、C(假手术2周组)、D(假手术4周组)4组,每组6只,放免法测定各组雌性大鼠血清雌二醇水平,运用免疫组化和RT-PCR对HO-1、HO-2在阴道平滑肌进行定位和半定量分析。结果:术后A与C、B与D组血清雌二醇比较有显著差异(P<0.05);HO-1、HO-2免疫组化定位在阴道平滑肌细胞胞质内,A组表达显著低于C组;阴道平滑肌组织中HO-1、HO-2mRNA表达在A、B组均显著低于C、D组,阴道中HO-1mRNA、HO-2mRNA下降与血清雌二醇下降相关。结论:HO-1、HO-2在雌性大鼠阴道平滑肌中有表达,低水平的雌激素与HO-1、HO-2表达降低有关,雌激素可能经HO/CO途径在阴道平滑肌中发挥生理作用。
Objective: To investigate the expression of HO-1 and HO-2 in vaginal smooth muscle of ovariectomized female rats and to explore the effect of estrogen on the expression of HO-1, HO- 2 expression. Methods: Female 8-week-old SD rats were randomly divided into 4 groups: A (ovariectomized for 2 weeks), B (ovariectomized for 4 weeks), C (sham operated for 2 weeks), D Six rats in each group were randomly divided into three groups. The serum estradiol level was determined by radioimmunoassay. The localization and semi-quantitative analysis of HO-1 and HO-2 in vaginal smooth muscle were performed by immunohistochemistry and RT-PCR. Results: There was significant difference in serum estradiol between A and C, B and D groups (P <0.05). The expression of HO-1 and HO-2 in the cytoplasm of vaginal smooth muscle cells was significantly lower than that in group A In group C, the expression of HO-1 and HO-2mRNA in vaginal smooth muscle was significantly lower in group A and B than in group C and D, and the decrease of HO-1mRNA and HO-2mRNA in vagina was associated with the decline of serum estradiol. CONCLUSION: HO-1 and HO-2 are expressed in the vaginal smooth muscle of female rats. Low estrogen levels are associated with decreased expression of HO-1 and HO-2. Estrogen may play a physiological role in vaginal smooth muscle via HO / CO pathway effect.
