目的研究载有多种癌变相关基因的17号染色体在不同器官、不同组织类型肿瘤单个细胞内拷贝数的变化规律。方法以正常外周血淋巴细胞为对照,采用FISH方法检测各细胞株17号染色体拷贝数。结果17号染色体在各细胞株单个细胞中拷贝数表现为:SKOV3以六倍体为主,占48%,七倍体率占23%;3AO以三倍体为主,占93%;OV2以四倍体为主,占67%;MGC803以六倍体为主,占64%;BGC823以二倍体为主,占83%;SGC7901以三倍体为主,占80%;PAMC82以五倍体为主,占78%。结论各细胞系均无17号染色体缺失,反呈扩增趋势。
Objective To study the variation of the copy number of chromosome 17 in different organs and different tissue types of chromosome 17 carrying a variety of cancer-associated genes. Methods Normal peripheral blood lymphocytes were used as controls. FISH was used to detect chromosome 17 copies of each cell line. RESULTS: The copy number of chromosome 17 in individual cell lines was: SKOV3 was predominantly hexaploid, accounting for 48%, and heptaploid rate was 23%; 3AO was mainly triploid, accounting for 93%; OV2 was Tetraploid predominates, accounting for 67%; MGC803 is predominantly hexaploid, accounting for 64%; BGC823 is predominantly diploid, accounting for 83%; SGC7901 is predominantly triploid, accounting for 80%; PAMC82 is fivefold The main body, accounting for 78%. Conclusion There is no deletion of chromosome 17 in all cell lines.
