目的观察对槲皮素人慢性粒细胞白血病细胞株K562增殖的影响。方法不同浓度槲皮素孵育K562细胞48小时后用CCK-8法检测细胞存活率;Western-blot法检测槲皮素孵育K562 p-AMPK(磷酸腺苷活性激酶)和Cox-2(环氧化酶-2)蛋白表达水平。结果槲皮素抑制K562细胞的增殖,呈时间和浓度依赖性。槲皮素孵育48h后K562细胞的IC50值是83.11μm;随着槲皮素浓度的增加,K562细胞Cox-2蛋白水平表达下降,而p-AMPK蛋白表达水平上升。结论槲皮素呈浓度依赖性抑制HL-60细胞的增殖,可能通过激活AMPK抑制Cox-2进而抑制K562细胞增殖。
Objective To observe the effect on the proliferation of quercetin human chronic myeloid leukemia cell line K562. Methods K562 cells were incubated with different concentrations of quercetin for 48 hours and cell viability was determined by CCK-8 assay. K562 p-AMPK (phosphorylated adenosine kinase activity) and Cox-2 Enzyme-2) protein expression level. Results Quercetin inhibited the proliferation of K562 cells in a time and concentration-dependent manner. The IC50 value of K562 cells after quercetin incubation for 48 h was 83.11 μm. With the increase of quercetin concentration, the expression of Cox-2 protein decreased and the expression of p-AMPK protein increased in K562 cells. Conclusions Quercetin inhibits the proliferation of HL-60 cells in a concentration-dependent manner, and may inhibit the proliferation of K562 cells by inhibiting the expression of Cox-2 by AMPK.
