目的 研究在药物诱导小鼠红白血病细胞系EML沿红系分化过程中基因表达的变化。方法 用mRNA差异显示法分析二甲亚砜 (DMSO)和血红素 (Hemin)诱导MEL细胞分化前后基因表达的改变 ,并利用改进的cDNA末端快速扩增 (RACE)技术克隆其中一个差示cDNA片段的旁侧序列 ;以Northern杂交分析检测对应基因的表达改变。结果 在差异显示分析中检测到长 115bp的转录本在诱导后表达增强。经RACE扩增后产物长达 1883bp ,和小鼠BVL 1转座成分的内部序列 (195 5~ 40 0 0nt)区段有很高的同源性。Northern杂交表明MEL细胞中该基因至少有 3个转录本 ,在诱导后表达水平均显著增高。结论 在MEL细胞红系分化过中发现了一种新的VL 30样基因。
Objective To study the changes of gene expression during drug-induced erythroid differentiation of mouse Erythroleukemic cell line EML. Methods mRNA differential display was used to analyze the changes of gene expression before and after MEL cell differentiation induced by dimethyl sulfoxide (DMSO) and hemin (Hemin), and one of the differentially expressed cDNA fragments was cloned by modified rapid amplification of cDNA ends (RACE) technique. The flanking sequence; Northern blot analysis to detect changes in the expression of the corresponding gene. Results Increased expression of the long 115 bp transcript was detected in differential display analysis. The product amplified by RACE was as long as 1883 bp and had high homology with the internal sequence (195 5-400 nt) of the mouse BVL 1 transposition component. Northern hybridization showed that there were at least 3 transcripts of this gene in MEL cells, and the expression level was significantly increased after induction. Conclusion A new VL 30-like gene was found in the erythroid differentiation of MEL cells.
