目的研究MTLC(cmyc target from laryngeal cancercells)基因在喉癌组织中的表达规律及其在喉癌发生中的作用。方法用反转录聚合酶链反应法检测MTLCmRNA在喉癌组织及配对对照组织中的表达水平;用基因转染分析MTLC基因过表达对Hep2细胞生长的影响。结果36份喉癌组织中有28份(77.8%)MTLCmRNA表达下调;与转染空质粒的对照细胞相比,表达MTLC的细胞生长明显受到抑制,凋亡细胞比例明显增高(喉癌组织为118/300,对照细胞为25/300)。结论MTLC基因在喉癌组织中表达下调,其过表达可抑制喉癌Hep2细胞生长并促进凋亡,提示MTLC可能在喉癌发生、发展过程中起重要作用。
Objective To study the expression of MTLC (cmyc target from laryngeal cancer cells) in laryngeal carcinoma and its role in the development of laryngeal carcinoma. Methods The expression of MTLC mRNA in laryngeal carcinoma and matched control tissues was detected by reverse transcription polymerase chain reaction (RT-PCR). The effect of MTLC gene overexpression on the growth of Hep2 cells was analyzed by gene transfection. Results Twenty-eight (77.8%) MTLC mRNA expressions were down-regulated in 36 laryngeal cancer tissues. Compared with the control cells transfected with empty plasmid, MTLC-expressing cells were significantly inhibited and the percentage of apoptotic cells was significantly increased / 300, control cells 25/300). Conclusion The MTLC gene is down-regulated in laryngeal squamous cell carcinoma. Overexpression of MTLC gene can inhibit the growth of Hep2 laryngeal carcinoma cells and promote apoptosis. It suggests that MTLC may play an important role in the occurrence and development of laryngeal carcinoma.
