目的检测汽油机尾气对大鼠慢性肺损伤及不同类型胶原蛋白表达的影响。方法将健康SPF级雄性大鼠30只随机分为6组,即对照组和染毒1、2、4、8、12周组,每组5只,将汽油机尾气通入装有实验大鼠的自制染毒容器中染毒,每天染毒2 h,分别于第1、2、4、8、12周观察肺组织病理学变化,并用免疫组织化学技术检测CollagenⅠ、CollagenⅢ、CollagenⅣ胶原蛋白。结果 Collagen I、CollagenⅢ主要分布于支气管周围与血管周围,阳性表达强弱和面积随染毒时间的延长而增加。CollagenⅣ的表达在染毒2周内与正常对照组无明显差异;但染毒4周后其表达因部位不同而异,肺间隔随染毒时间延长而增厚,肺泡结构破坏,肺泡上皮基底膜的阳性表达面积逐渐减小;但支气管与血管周围阳性表达的强度与面积随染毒时间的延长而增加。结论 CollagenⅠ、CollagenⅢ、CollagenⅣ是汽油机尾气造成大鼠慢性肺损伤时表达的主要胶原蛋白。
Objective To detect the effects of gasoline tail gas on chronic lung injury and the expression of different types of collagen in rats. Methods Thirty healthy SPF male rats were randomly divided into 6 groups, namely control group and 1, 2, 4, 8 and 12 week groups, with 5 rats in each group. The tail gas of gasoline engine was introduced into experimental rats The cells were exposed to homemade containers for infection for 2 hours. The pathological changes of the lungs were observed on the 1st, 2nd, 4th, 8th and 12th weeks respectively. Collagen Ⅰ, Collagen Ⅲ and Collagen Ⅳ were detected by immunohistochemistry. Results Collagen I and Collagen III mainly distributed around the bronchus and perivascular area, and the positive expression intensity and area increased with the extension of exposure time. The expression of CollagenⅣwas not significantly different from that of the normal control group within 2 weeks after exposure. However, the expression of CollagenⅣwas different from that of the normal control group at 4 weeks after exposure. The interval between lungs increased with the prolongation of exposure time, the destruction of alveolar structure, The area of positive expression decreased gradually; however, the intensity and area of positive expression around the bronchus and perivascular increased with the extension of exposure time. Conclusions Collagen Ⅰ, Collagen Ⅲ and Collagen Ⅳ are the main collagen expressed in rats with chronic lung injury induced by exhaust gas of gasoline engine.
