本试验应用同工酶的聚丙烯酰胺凝胶垂直平板电泳分析,对42个二棱大麦(Hordeum disticum L.)品种进行了品种鉴定。根据干种子、脏、胚乳、叶片、芽鞘和根的酯酶同工酶电泳图谱类型,可将26个品种逐个鉴定出来,占品种总数的61.9%。根据胚、胚乳和叶片的过氧化物酶同工酶电泳图谱差异,可将16个品种逐个鉴定出来,占品种总数的38.1%。根据胚的多酚氧化酶同工酶电泳图谱差异,可将供试品种分成4组。根据干种子、胚和胚乳的α—淀粉酶同工酶电泳图谱类型,可将供试品种分成18个组,其中,11个品种可逐个地鉴定出来。这种方法具有鉴别效果好,操作技术不很复杂,所需仪器设备比较普通,并且费用低、速度快等特点,是鉴定二棱大麦品种的有效手段。因此,在生产实践中值得应用推广。
In this experiment, 42 isolates of Hordeum disticum (L.) were identified by isoenzyme polyacrylamide gel electrophoresis. According to the types of esterase isoenzyme electrophoresis map of dry seeds, dirty, endosperm, leaves, bud sheath and root, 26 varieties can be identified one by one, accounting for 61.9% of the total varieties. According to the differences of peroxidase isozyme electrophoresis patterns of embryos, endosperm and leaves, 16 varieties can be identified one by one, accounting for 38.1% of the total varieties. According to embryo polyphenol oxidase isozyme electrophoresis differences, the test varieties can be divided into 4 groups. According to the types of α-amylase isozymes of dry seeds, embryos and endosperm, the tested varieties can be divided into 18 groups, of which 11 varieties can be identified one by one. The method has the advantages of good identification effect, not very complicated operation technique, relatively common required equipment and equipment, low cost and high speed, and is an effective method for identifying two-rowed barley varieties. Therefore, it is worth to apply the promotion in the production practice.
